1750 CUNICAL CHEMISTRY, Vol. 39, No. 8, 1993 pam, oxazepam, triazolam) or the antidepressants amitriptyline, clomipramine, demethyl domipramine, desipramine, imipramine, fluvoxamine, trimipramine; nortriptyline, however, interferes with norfluoxetine. Blood samples from patients treated with a combination of fluoxetine and benzodiazepines (bromazepam, alprazolam, temazepam, clorazepate dipotassium, oxazepam) were also tested and showedno interferences. For nine patients whose daily dose of fluoxetine or norfluoxetine was between 20 and 40 mg, blood concentrationsobtained were 24 to 420 ugfL and 52 to 419 igfL, respectively. These ranges are similar to those obtained in previous studies (5,10). Our results clearly show that (a) a blood concentrationof 10 gIL for fluoxetine or norfluoxetine can be detected by the described method, and (b) relevant interferences noted in other studies, especially benzodiazepines and imipramine, are not a problem in this method.

Measurement of urinary oxalate by grain sorghum leaf oxalate oxidase immobilized to affixed alkylamine glass beads.

Oxalate in urine was measured by grain Sorghum leaf oxalate oxidase conjugated to alkyl amine glass beads affixed in a beaker. The minimum detection limit was 0.05 mM/L in urine. Recovery of added oxalate in urine was 80.5% and within and between assay, coefficients of variation (CV) were <4% and <5.5%, respectively. Urinary oxalate values obtained by the present method showed a good correlatio...

Molecular characterization of the oxalate oxidase involved in the response of barley to the powdery mildew fungus.

Previously we reported that oxalate oxidase activity increases in extracts of barley (Hordeum vulgare) leaves in response to the powdery mildew fungus (Blumeria [syn. Erysiphe] graminis f.sp. hordei) and proposed this as a source of H2O2 during plant-pathogen interactions. In this paper we show that the N terminus of the major pathogen-response oxalate oxidase has a high degree of sequence iden...

Barley (Hordeum vulgare) oxalate oxidase is a manganese-containing enzyme.

Oxalate oxidase (EC 1.2.3.4) catalyses the conversion of oxalate and dioxygen into CO(2) and H(2)O(2). The barley (Hordeum vulgare) seedling root enzyme was purified to homogeneity and shown by metal analysis and EPR spectroscopy to contain Mn(II) at up to 0.80 atom per subunit. The involvement of Mn and neither flavin, Cu nor Fe in the direct conversion of dioxygen to H(2)O(2) makes oxalate ox...

Assay of plasma oxalate with soluble oxalate oxidase.

We use oxalate oxidase from barley seedlings for the colorimetric determination of oxalate in plasma. The oxalate is converted to hydrogen peroxide, which, in the presence of peroxidase, is detected by a Trinder-like chromogenic system. Optimization of the assay, including deproteinization and elimination of interferences from reducing substrates, is described. Ascorbate additions (200 mumol/L)...

Urinary & serum oxalate determination by oxalate oxidase immobilized on to affixed arylamine glass beads.

BACKGROUND & OBJECTIVE Although the measurement of oxalate in urine and serum by Amaranthus leaf oxalate oxidase immobilized on free arylamine glass beads is highly sensitive and specific, the handling of glass beads is tedious and cumbersome. The present study was undertaken to overcome this problem. METHODS Partially purified Amaranthus spinosus leaf oxalate oxidase was immobilized through ...
